摘要:目的:比较两种检测方法优劣,为丰富山银花药材质量控制方法提供参考。方法:采用ACQUITY UPLC BEH C18(2.1×100mm,1.7μm)色谱柱,流动相乙腈(A)-0.4%醋酸(B),梯度洗脱(0~2 min,7%A;2~2.3 min,7%A^28%A;2.3~10 min,28%A;10~10.5 min,28%A^7%A),梯度流速(0~2~2.3~2.6~2.8~4~4.3~4.5~10.5min,0.35~0.35~0.25~0.15~0.1~0.2~0.3~0.35~0.35ml/min),PDA(二极管阵列检测器)λ=330 nm;ELSD检测器,漂移管温度60℃,喷雾器48℃,增益值100,气体流速20psi,柱温50℃。结果:PDA检测绿原酸在0.3045~1.5225μg(R2=0.9994)质量范围内线性关系良好,ELSD检测绿原酸在0.3045~1.5225μg(R2=0.9993)质量范围内线性关系良好。结论:PDA检测器检测时受噪音影响较小,基线较平稳,测得的数据比ELSD检测器更准确。
Objective Compare two methods of testing,and which one was better.Provide a reference for rich Flos Lonicera me-dicinal materials quality control method .Methods ACQUITY UPLC BEH C18 (2.1×100 mm,1.7μm)column was adopted and the temperature was 50 ℃.Two solution were acetonitrile (A)and 0.4% acetic acid (B),with the gradient elution of (0 -2 min,A 7%;2 -2.3 min,A 7% -28%;A 2.3 -10 min,A 28%;10 -10.5 min,A 28% -A7%),and velocity gradient (0 -2-2.3 -2.6 -2.8 -4 -4.3 -4.5 -10.5 min,0.35 -0.35 -0.25 -0.15 -0.1 -0.2 -0.3 -0.35 -0.35 ml/min).The detection waveleth was 330 nm .Electronic Levelling Suspension ,The temperature of drift tube was maintained at 60℃, the temperature of nebulizer was 48℃,PMT was 100.with the nitrogen flow rate of 20 psi and column temprerature was 50℃. Results The linear response ranges of Chlorogenic Acid was 0.3045 -1.5225μg (R^2 =0.9994)with PDA and 0.3045 -1.5225μg (R^2 =0.9993)with ELSD respectively.Conclusion Photo -diode array was less about noise,more smooth about base line and more accurate about date than Evaporative Light -scattering Detector.
