摘要:目的:研究补脾强力方对实验性自身免疫重症肌无力大鼠的治疗作用及量-效关系。方法:选择Lewis大鼠,经造模,随机分为正常对照组、模型对照组、强的松5.4 mg/kg组、补脾强力方9.49 g/kg、7.12 g/kg、4.75 g/kg组,除正常对照组外,均采用鼠源性AchR-α亚基97-116肽段序列(R97-116)免疫接种复制实验性自身免疫重症肌无力大鼠模型。给药后,采用ELISA法检测各组大鼠血清中AchR-Ab水平及细胞因子IFN-γ、TNF-α、IL-4、IL-10、IL-17的含量。结果:补脾强力方对实验性自身免疫重症肌无力大鼠体质量的作用效果与给药剂量的高低成正相关;与模型对照组相比,强的松组5.4 mg/kg组、补脾强力方9.49 g/kg、7.12 g/kg、4.75 g/kg组中AchR-Ab水平及细胞因子IFN-γ、TNF-α、IL-4、IL-17的含量明显下降,且下调趋势与剂量高低成正相关。而大鼠血清中IL-10的含量,各给药组间均无明显差异。结论:补脾强力方可明显改善实验性自身免疫重症肌无力大鼠的体质量,同时,可明显下调该模型大鼠血清中细胞因子IFN-γ、TNF-α、IL-4、IL-17的含量,抑制AchR-Ab的产生而起到治疗作用,且干预效果与给药剂量成正相关。
Objective: To study the therapeutic effect and dose-effect relationship of Bupiqiangli compound on experimental autoimmune myasthenia gravis of rats. Methods: Lewis rats were randomly divided into the control group,the model group,5. 4 mg/kg prednisone group,9. 49 g/kg,7. 12 g/kg,4. 75 g/kg Bupiqiangli compound groups,all Lewis rats were immunized with 97-116 peptide of the rat AchR-α subunit(R97-116) to copy experimental autoimmune myasthenia gravis rats model except the control group. After the treatment,contents of serum AchR-Ab levels and cytokines IFN-γ,TNF-α,IL-4,IL-10,IL-17 were detected by ELISA method. Results: There was the positive correlation beween the dose and the effect of Bupiqiangli compound on the body quality of experimental autoimmune myasthenia gravis rats. Compared with the model group,AchR-Ab level and cytokines IFN-γ,TNF-α,IL-4,IL-17 obviously decreased in 5. 4 mg/kg prednisone group,9. 49 g/kg,7. 12 g/kg,4. 75 g/kg Bupiqiangli compound groups(P〈0. 01),and the decline degree and the dose was closely relative.There was no obvious difference in serum IL-10 level among Bupiqiangli compound groups(P〈0. 05). Conclusion: Bupiqiangli compound can obviously improve the body quality of the experimental autoimmune myasthenia gravis rats,at the same time,obviously decrease serum cytokines IFN-γ,TNF-α,IL-4,IL-17 in autoimmune myasthenia gravis rats,and suppress the produce of AchR-Ab,and the intervention effect is positively related to the dose.
