摘要:研究了药食兼用植物夜寒苏粗多糖体外抗氧化及抑制α-葡萄糖苷酶的活性。采用闪式提取法提取夜寒苏粗多糖,并以苯酚-硫酸法测定多糖的提取率;测定夜寒苏粗多糖对DPPH自由基、ABTS^(+)自由基的清除能力及还原能力,以评价其体外抗氧化活性,并研究其对α-葡萄糖苷酶活性的影响。结果表明,夜寒苏多糖提取率为1.56%,多糖含量为63.23%,其清除DPPH自由基、ABTS+自由基的IC50值分别为55.21±1.05 mg/m L,6.09±0.33 mg/m L,对Fe^(3+)的还原能力为(0.66±0.08 mmol FeSO4)/g,抗氧化活性随浓度的增高而增强,但活性弱于维生素C(Vc);夜寒苏粗多糖具有抑制α-葡萄糖苷酶活性,且表现浓度依赖性。研究表明,夜寒苏粗多糖具有体外抗氧化活性和抑制α-葡萄糖苷酶活性,为开发夜寒苏的食用、药用价值提供理论依据。
The aim of this study was to study the antioxidant activity and inhibition ofα-glucosidase activity in vitro of the crude polysaccharides from the medical and edible plant Hedychium flavum Roxb.The crude polysaccharides was extracted from Hedychium flavum Roxb.by flash extraction,and the extraction rate was determined by phenol-sulfuric acid colorimetry assay.The antioxidant activity of the crude polysaccharides in vitro was determined by DPPH,ABTS^(+)free radical scavenging,and Fe^(3+ )reducing assays,and the effect onα-glucoside activity was studied.The results showed that the extraction rate of polysaccharides was 1.56%,the purity of polysaccharides was 63.23%,the IC50values of scavenging DPPH and ABTS+free radical were 55.21±1.05 mg/m L and 6.09±0.33 mg/m L,respectively.The Fe3+-reducing capacity was 0.66±0.08 mmol FeSO4/g.The antioxidant activity increased with the increase of concentration,but the activity was weaker than vitamin C.At the same time,the crude polysaccharides showedα-glucosidase inhibitory activity in a concentration dependent manner.This study exhibited that the crude polysaccharides from Hedychium flavum Roxb.had antioxidant activity and inhibitory activity againstα-glucosidase,which provided theoretical basis for the development of edible and medicinal value of Hedychium flavum Roxb.
