摘要:通过闪式提取、分级醇沉制备分级多糖;通过红外、紫外等实验测定分级多糖理化性质;通过清除DPPH自由基、羟自由基实验评估分级产物抗氧化活性;通过抑制α-淀粉酶实验评估分级产物降血糖活性;通过旋转式黏度计测定不同温度下多糖黏度变化。结果表明,DOP60多糖含量较高、活性较好,可作为铁皮石斛应用于护肤、特医食品的二次开发工业原料。在高于50℃时,多糖黏度较低,变化平缓,易于过滤,解决了在生产中多糖水提液与石斛粉渣难以分离问题。
The fractionated polysaccharide was prepared from dendrobium officinale by flash extraction and fractional alcohol precipitation;the physicochemical properties of the fractionated polysaccharide were determined by infrared and ultraviolet experiments;the antioxidant activity of the fractionated product was evaluated by scavenging DPPH free radicals and hydroxyl free radicals;the inhibition ofα-amylase was tested for evaluating the hypoglycemic activity.The viscosity change of polysaccharides at different temperatures was measured by rotational viscometer.The results show that DOP60 polysaccharide content is higher and the activity is better,which can be used as the secondary-development industrial raw material from dendrobium officinale for application in the fields of skin care and special medical food.When the temperature is higher than 50℃,the viscosity of polysaccharide is low,the change in viscosity is gentle,and it is easy to be filtered,which solves the difficulty in separation between polysaccharide aqueous extract and dendrobium residue powder in production.The method is simple and effective,and has the potential of industrial application.
