摘要:为提取分离响铃草中黄酮类成分,探索响铃草中黄酮类成分的抗氧化能力。在单因素试验基础上,以总黄酮含量为因变量,选择提取时间、乙醇浓度和料液比为影响因素,采用Box-Behnken响应面法优化响铃草总黄酮的提取工艺,以最优方案提取的总黄酮进行DPPH、还原力和羟自由基的抗氧化性试验。响应面设计后最优的提取工艺为提取时间92 s,乙醇浓度60 %,料液比1 ∶ 35(g/mL),响铃草总黄酮含量为21.414 2 mg/g 。体外抗氧化性试验显示总黄酮具有较高的清除自由基能力和较高的还原力,且与总黄酮含量呈量效关系。该方法稳定可靠,可用于响铃草总黄酮的提取和抗氧化性测定,为响铃草的进一步开发和应用提供一定依据。
In order to extract and isolate the flavonoids from Crotalaria ferruginea, the antioxidant ability of flavonoids was studied. On the basis of single-factor test,the content of total flavonoids was used as the responsive values, extraction time,ethanol concentration and material-to-liquid ratios were set as the main influencing factors, response surface methodology were used to optimize the extraction process of total flavonoids from Crotalaria ferruginea. The antioxidant capacity of total flavonoids from optimal extraction method was studied by DPPH free radical,reducing power and H2O2 free radical scavenging activity. The optimal extraction conditions of total flavonoids by response surface methodology were extraction time of 92 second,ethanol concentration of 60 % and material-liquid ratio of 1 ∶ 35(g/mL). The content of flavonoids was 21.414 2 mg/g. Results of antioxidation showed that the total flavonoids had a good ability to scavenge radicals and had good antioxidant activity in a dose-dependent manner. The stable and reliable method was used for the extraction and antioxidant determination of total flavonoids of Crotalaria ferruginea,which provides some basis for the further development and application of Crotalaria ferruginea.
