摘要:目的 研究水芹乙醇提取物的体外抗炎活性,并探讨其作用机制。方法 采用回流提取法制备水芹乙醇提取物,通过可见分光光度法测定其总黄酮和总酚含量,采用超高效液相色谱-四极杆-静电场轨道阱串联质谱(UPLC-Q-Orbitrap MS)技术鉴定其化学成分。利用脂多糖(LPS)诱导RAW264.7细胞炎症模型,在药物干预24 h后,检测细胞上清液中一氧化氮(NO)、白细胞介素1β(IL-1β)、白细胞介素6(IL-6)和肿瘤坏死因子α(TNF-α)等炎症因子的释放量;通过DCFH-DA荧光探针法测定细胞中的活性氧(ROS)水平;采用RT-qPCR检测诱导型一氧化氮合酶(iNOS)、环氧化酶-2(COX-2)、TNF-α、IL-6和IL-1β的mRNA表达水平,Western blot法检测iNOS、COX-2、丝裂原活化蛋白激酶(MAPK)和核转录因子-κB(NF-κB)信号通路相关蛋白的表达。结果 水芹乙醇提取物中总黄酮含量为6.73%,总酚含量为4.47%。共鉴定出51个化学成分,包括7种黄酮类和11种酚类化合物。水芹乙醇提取物能够显著抑制NO、TNF-α、IL-6和IL-1β的释放(P<0.05或P<0.01),降低细胞内ROS水平,并下调iNOS、COX-2、TNF-α、IL-6和IL-1β的mRNA表达(P<0.05或P<0.01)。同时,水芹乙醇提取物还能够下调iNOS、COX-2、细胞核NF-κB p65蛋白的表达,以及丝裂原活化蛋白激酶(p38)、应激活化蛋白激酶(JNK)、细胞外调节蛋白激酶(ERK)和核因子κB抑制蛋白(IκBα)蛋白的磷酸化水平(P<0.05或P<0.01)。结论 水芹乙醇提取物能够抑制LPS诱导的RAW264.7细胞炎症反应,其机制与MAPK和NF-κB信号通路的调控有关。
OBJECTIVE To investigate the in vitro anti-inflammatory effects of ethanol extract from Oenanthe javanica and elucidate its underlying mechanisms.METHODS The ethanol extract of Oenanthe javanica was prepared using reflux extraction,with total flavonoid and phenolic contents measured by visible spectrophotometry.The chemical components of the extract were identified by ultra-high performance liquid chromatography-quadrupole-orbitrap mass spectrometry(UPLC-Q-Orbitrap MS).An inflammation model was established by inducing RAW264.7 cells with lipopolysaccharide(LPS).After 24 hours of treatment with the extract,the release levels of nitric oxide(NO),interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)in the cell supernatant were determined.Intracellular reactive oxygen species(ROS)levels were evaluated with the DCFH-DA fluorescent probe method.Additionally,RT-qPCR was used to evaluate the mRNA expression levels of iNOS,COX-2,TNF-α,IL-6,and IL-1β.Western blotting analysis was conducted to detect the expression of iNOS,COX-2 proteins,and MAPK and NF-κB pathway-related proteins.RESULTS The extract of Oenanthe javanica contained 6.73%flavonoids and 4.47%phenolics.A total of 51 chemical components were identified,including 7 flavonoids and 11 phenols.The extract significantly inhibited the release of NO,TNF-α,IL-6,and IL-1β(P<0.05 or P<0.01),reduced intracellular ROS levels,and downregulated the mRNA expression of iNOS,COX-2,TNF-α,IL-6,and IL-1β(P<0.05 or P<0.01).Additionally,the extract decreased the protein expression of iNOS,COX-2,nuclear NF-κB p65,and phosphorylated p38,JNK,ERK,and IκBα(P<0.05 or P<0.01).CONCLUSION The ethanol extract of Oenanthe javanica inhibits the LPS-induced inflammatory response in RAW264.7 cells,and its mechanism is associated with the regulation of the MAPK and NF-κB signaling pathways.
