摘要:为探讨生长素对荠菜心皮发育的影响,构建了生长素报告系统DR5∶∶GFP,通过农杆菌介导法获得了转基因荠菜植株。荧光显微镜观察结果显示,荠菜心皮发育早期上端两侧荧光信号较其他部分强。为进一步探讨其分子机制,以RT-PCR方法克隆了荠菜PIN3基因(CbPIN3),并做了生物信息学分析。结果表明,CbPIN3基因编码区全长1 950 bp,可编码649个氨基酸。CbPIN3蛋白存在13个丝氨酸磷酸化位点,包含9个跨膜结构域,与拟南芥PIN3亲缘关系最近。此外,实时荧光定量PCR(qRT-PCR)方法检测荠菜CbPIN3组织表达特点,发现CbPIN3基因在荠菜多种组织中均表达,在茎中表达量最高,果荚中表达量最低。
To probe the effects of auxin on the carpel development of Capsella bursa-pastoris,auxin reporting system DR 5∶∶GFP was constructed,and then transgenic Capsella bursa-pastoris seedling was obtained via Agrobacterium-mediated transformation.Results demonstrated that the fluorescence signals were stronger in both upper sides than middle and base sides of the carpel.PIN 3(CbPIN 3)gene coding sequences were cloned by RT-PCR to further explore the molecular mechanism.Sequence analysis showed that the length of the CbPIN 3 gene cDNA was 1 950 bp encoding 649 amino acids.Information analysis indicated that CbPIN3 had 13 serine phosphorylation sites,included 9 transmembrane domains,and had the closest affinity with PIN3 of Arabidopsis thaliana.In addition,qRT-PCR results suggested that CbPIN 3 expressed in all tissues detected,and the highest expression site was stem,and the lowest site was silique.
