摘要:目的:对贵州省各产地何首乌中卵磷脂成分进行分析,建立了何首乌卵磷脂的含量测定方法,为后期该药材用于功能性食品和保健品等产品的开发和利用提供理论依据。方法:采用高效液相色谱法,运用蒸发光散色检测器进行检测,色谱柱为Angilent SIL柱(5μm,4.6mm×250mm),以甲醇-水-冰醋酸-三乙胺(85∶15∶0.45∶0.05)为流动相A,以正己烷-异丙醇-流动相A(20∶48∶32)为流动相B,梯度洗脱,柱温40℃;流速为1mL/min;进样量为10μL,漂移管温度为90℃;载气流量为2.8L/min,记录时间为20min。结果:测定了16批贵州不同产地的何首乌卵磷脂的含量,平均含量为1.98%,并且生长年限越长,其卵磷脂含量越高。结论:所建立的方法简便、可靠,可作为何首乌中功能性成分卵磷脂的检测,为后期产品的开发和利用提供技术支持。
Objective:The analysis of phospholipid content in Polygonum multiflorum Thunb from different producing areas in Guizhou Province was carried out,and the method for determining the content of phospholipid in Polygonum multiflorum Thunb was established,which provided a theoretical basis for the development and utilization of this medicine for functional foods and health products.Method:High performance liquid chromatography,using the ELSD to detect the sample.Chromatographic column: Angilent SIL column(5μm,4.6 mmx250 mm).Mobile fhase A:methanol-water-glacialaceticacid-triethylamine(85∶15∶0.45∶0.05).Mobile phase B: Nhexane-isopropanol-mobile phase A(20∶48∶32),gradient elution.Column temperature: 40 degree centigrade.Velocity: 1 mL/min.Injection volume:10 μL.Drifit tube temperature:90 degree centigrade.Carrier gas flow rate:2.8 L/min.Analysis time: 20 min.Results: Determination of the phospholipid content of Polygonum multiflorum Thunb in 16 batches from different regions of Guizhou,the average content is 1.98%,and the longer the growth period,the higher the phospholipid content.Conclusion:The established method is simple and reliable,and can be used as a detection of phospholipid,a functional component in Polygonum multiflorum Thunb,and provides technical support for the development and utilization of later products.
