摘要:目的研究锦鸡儿总黄酮(TFC)对大鼠局灶性脑缺血再灌注损伤后缺血周边区星形胶质细胞胶质纤维酸性蛋白(GFAP)生长的影响。方法将大鼠随机分为假手术组、模型组、TFC高、中、低剂量(60、30、15 mg/kg)组,采用线栓法制作大鼠局灶性脑缺血再灌注损伤模型,脑缺血1.5 h再灌注3 h后,TFC高、中、低剂量组分别给予TFC 60、30、15 mg/kg灌胃治疗,假手术组及模型组给予等体积生理盐水,每日1次,连续治疗14 d,观察各组大鼠在脑缺血后24 h和治疗第14天后神经功能缺损综合评分,苏木素-伊红(HE)染色观察缺血周边区脑组织形态结构的变化,荧光免疫组织化学染色及Western印迹法检测各组脑缺血边缘区GFAP表达水平。结果脑缺血24 h后,与模型组比较,TFC高、中、低剂量组神经功能缺损评分差异无统计学意义(P>0.05);脑缺血14 d后,与模型组比较,TFC高、中、低剂量组神经功能缺损评分均显著降低(P<0.05,P<0.01),TFC高、中、低剂量组均明显促进脑缺血边缘区神经细胞的修复;模型组和TFC高、中、低剂量组GFAP阳性细胞数及蛋白表达量均显著高于假手术组(P<0.01),TFC高、中剂量组GFAP阳性细胞数及蛋白表达量显著少于模型组(P<0.05,P<0.01),细胞生长形态规则。结论 TFC能改善脑缺血后神经功能缺损症状,可促进神经细胞恢复,其机制可能与抑制缺血半暗带区GFAP的过度生长有关。
Objective To study the effect of total flavonoids of Caraganamicrophylla on the expression of GFAP in the penumbra of focal cerebral ischemia-reperfusion injury in rats. Methods Rat model of cerebral ischemia-reperfusion injury was created by the middle cerebral artery occlusion (MCAO). The SD rats (250-300 g, male) were randomly divided into five groups: sham, model and total chicken flavonoids (60, 30, 15 mg/kg) groups. Drugs were administered intragastrically once a day for 14 days starting from 24 h after reperfusion, and the rats in the sham group and model group were given equal volume of normal saline. The HE staining for morphological changes and neurological deficit score were performed after MCAO. The expression of GFAP in penumbra was investigated by immunohistochemically staining and Western blot 14 days after MCAO. Results Compared with the sham group, MCAO could significantly increase the expression of GFAP(P〈0.01 ). Compared with the model group, treatment with Caragana total flavonoids significantly decreased neurological score( P〈 0. 05 ,P〈0.01 ), alleviated pathological damage and inhibited the expression of GFAP in penumbra at 14 d after cerebral ischemia-reperfusion( P〈0.05 ). However, Caragana total flavonoids could not significantly decrease neurological deficit at 12 h after cerebral ischemia-reperfusion ( P〉0.05 ). Conclusions The total flavonoids of Caragana could promote the recovery of brain cells and improve the neurological deficits in the rats with cerebral ischemia reperfusion, which might be associated with inhibiting the expression of GFAP in the ischemic penumbra.
