摘要:Endophytic fungi in karst extreme habitats serve as critical reservoirs for novel drug discovery;however, their diversity within medicinal ferns in karst ecosystems remains poorly characterized. Medicinal ferns in Guizhou, shaped by unique karst microhabitats(e.g., caves, fissures, and gullies), are hypothesized to harbor abundant untapped fungal resources. This study comprehensively characterizes the diversity, bioactivity potential, and secondary metabolite profiles of endophytic fungi associated with Dicranopteris ferns from Guizhou, with a specific emphasis on isolating and identifying secondary metabolites from selected strains. Key findings are as follows: 1) A total of 639 endophytic fungal strains were isolated from three Dicranopteris species viz. D. pedata, D. linearis, and D. obtusa across Guizhou Province, significantly expanding the microbial germplasm resources of Southwest China. 2) 431 species were identified from 639 strains, belonging to 4 phyla, 11 classes, 41 orders, 74 families, and 155 genera, including one new family, Massicellisporaceae, three new genera viz. Massicellispora, Miniglobosispora, and Neopopulomyces, 29 new species, and 36 new recorded species. 3) Secondary metabolites of 15 Dicranopteris endophytic fungi were analyzed. Seven strains with high secondary metabolite richness were selected, and 7 natural products were isolated, purified, and characterized using column chromatography combined with preparative chromatography. Among these, four were new structural compounds. This study not only enriches the fungal resource library of Southwest China but also provides critical resources and theoretical support for the development of endophytic fungal resources in Guizhou's medicinal ferns and the discovery of drug leads.
